A maioria dos estudos químicos de Protiuminvestigamprincipalmente os óleos essenciais extraídos da resina. No entanto, não há informações quanto à composição química do extrato das folhas, e quando não se dispõe de estudos químicos sobre a espécie,a análise fitoquímica pode identificar os grupos de metabólitos secundários relevantes. Este trabalho descreve a identificaçãodos metabólitos secundários do extrato etanólico obtido das folhas de Protium calendulinume a investigaçãodo seu potencial alelopático sobreLactuca sativa. As folhas de P.calendulinum foram coletadas em julho de 2019, na áreade vegetação da Universidade Federal do Amazonas-UFAM. Para a obtenção doextratoetanólico bruto (EB), foram adicionados 3 litros de álcool etílico 96%sobre 3 kg de folhas. A solução foi deixada em repouso por 15 diasesubmetida à destilação em rotaevaporador a 70° C. A prospecção fitoquímica doextrato EB foi realizada qualitativamente.Também foram feitas análises espectrofotométricas de varredura doEBnointervalo de 240 nm a 700 nm.Para o teste de alelopatia,o EBfoi diluído nas concentrações de125 mg/L-1, 250 mg/L-1, 500 mg/L-1e 1000 mg/L-1. Foi determinadaa condutividade elétrica das concentrações do extratoe os valores utilizados para determinar o potencial osmótico.Para o bioensaio dealelopatiaforamcolocadas 50 sementesdeL. sativaem placas de Petri, com papel filtro e adicionados 04 mLdas concentrações. Para o controle negativoutilizou-se água destilada.O potencial alelopático foi avaliadopor meio daporcentagem de germinação,Índice de Velocidade de Germinação (IVG),comprimento do hipocótilo e radícula. Posteriormente,determinou-se a concentraçãode clorofila e carotenoidesdas plântulas.Para avaliar o efeito das concentrações sobre porcentagem de germinação, IVG, comprimento do hipocótilo e da radículafoi utilizadaregressão linear,seguida da análise de variância (Teste F). Os resultados fitoquímicos do extrato de P. calendulinumevidenciaram a presença deflavonas, saponinas, catequinas, fenóis e antocianinas.Aliado aos resultados da prospecção fitoquímica, os dados da análise espectrofotométrica confirmarama presença de uma ampla variedade de metabólitos na faixa entre 240 a400 nm.Os resultadosdo potencial osmótico variaram entre -0,0003 MPae -0,0011 MPae estão de acordo com o limitede tolerância. Já para o potencialalelopático, foi observadoque com o aumento da concentração do extrato houvereduçãosignificativanagerminação, IVG, hipocótilo e radícula(p < 0,01). Tais efeitoscorroboramos resultados da prospecção fitoquímica, uma vez que, flavonas e saponinas em elevadas concentrações afetam a porcentagem de germinação, retardam o IVG e reduzem o crescimento do hipocótilo e radícula.Também houve redução dos teores de clorofila b nas plântulas conforme o aumento da concentraçãodo extrato EB. As sustâncias identificadas nesse estudo podem estar associadas aoelevado potencial alelopáticoapresentado peloextrato EBdas folhas de P. calendulinum,mesmo em baixas concentrações. Essas substânciaspodem contribuir para a caracterização e futuros estudos relacionados a essa espécie.

Most Protiumchemical studies mainly investigate essential oils extracted from the resin. However, there is no information regarding the chemical composition of the leaf extract, and when there are no chemical studies on the species, the phytochemical analysis can identify the relevant secondary metabolite groups. In this context, the aim of the study was to identify the secondary metabolites of the ethanolic extract, obtained from the leaves of Protiumcalendulinumand to investigate their allelopathic potential on Lactucasativa. P. calendulinumleaves werecollected in July 2019, in the vegetation area of the Federal University of Amazonas -UFAM. To obtain thecrude ethanolic extract (CEE), 3 liters of 96% ethyl alcohol on 3 kg of leaves were added. The solution was left to stand for 15 days and subjected to distillation in a rotary evaporator at 70° C. The phytochemical prospecting of the CEE as carriedout in a qualitative way. CEEspectrophotometricanalyzes were also performed, measured in the range of 240 nm to 700 nm. For the allelopathy test, EB was diluted in the following concentrations: 125 mg/L-1, 250 mg/L-1, 500 mg/L-1and 1000 mg/L-1. The electrical conductivity of the extract concentrations was determined, and the values used to determine the osmotic potential. The allelopathy bioassay was carried out on L. sativaseeds, 50 seeds were placed in Petri dishes and then added 04 mL of the concentrations. For the negative control, distilled water was used. The allelopathic potential was evaluated by means of the germination percentage, IVG (Germination Speed Index), length of the hypocotyl and radicle. After the test, the chlorophyll and carotenoid assay wereperformed. To assess the effect of concentrations on percentage ofgermination; IVG; length of the hypocotyl and length of the radicle, linear regression was used, followed by analysis of variance (Test F). The results obtained in the phytochemical prospecting of the extract of P. calendulinumshowed the presence of Flavones, Saponins, Catechins, Phenols and Anthocyanins. Coupled with the results of phytochemical prospecting, the spectrophotometric analysis data confirm the presence of a wide variety of metabolites in the rangebetween 240 and 400 nm. For the osmotic potential, the results obtained varied between -0,0003 MPa and -0,0011 MPa,and are in accordance with the tolerance limit. The results demonstrate that the increased concentration of the extract had a significant effect on germination, IVG, hypocotyl and radicle (p <0.01). This effect corroborates the results of phytochemical prospecting, since flavones and saponins in high concentrations affect the germination percentage, delay the speed with which the seeds germinate and reduce the growth of the hypocotyl and radicle. The levels of chlorophyll b decreased as the CEE concentrationincreased. The results obtained reveal the presence of different classes of secondary metabolites present in the extract of the leaves of P. calendulinum. And, possibly, due to the presence of these compounds, the extract has a high allelopathic potential, affecting the germination and initial growth of L.sativa, even in low concentrations.
